| I. Background | |
| CB1 受體（Cannabinoid Receptor Type 1）是內源性素系統（ECS）的核心成員， 屬于 G 蛋白偶聯受體（GPCR）家族，由基因 CNR1 編碼。CB1 廣泛分布于中樞神經系統 （如基底節、海馬、小腦）及外周組織（脂肪、肝臟、免疫細胞）。作為 脂質信號樞紐， CB1 通過結合內源性素（如 anandamide、2-AG）或外源性素（如Δ?-THC），調 控神經遞質釋放、突觸可塑性、能量代謝及疼痛感知。病理狀態下，CB1 過度激活與神經 精神疾?。ń箲]、成癮）、代謝紊亂（肥胖、胰島素抵抗）及慢性疼痛密切相關。盡管 CB1 拮抗劑利莫那班（Rimonabant）因精神副作用退市，新一代偏向性配體（如 CB1 負向變構調節劑）和靶向外周 CB1 的藥物正成為代謝病和神經退行性疾病的研究熱點。 | |
| II. Description | |
CB1 β-Arrestin CHO 報告基因藥靶模型很好的模擬了體內 CB1/β-Arrestin 的信號轉導過 程，原理見下圖所示。當缺乏配體刺激時，β-Arrestin 不與 CB1 結合，融合β-Arrestin 的熒 光素酶處于失活構象，當 CB1 遭遇配體刺激時，融合熒光素酶報告基因的β-Arrestin 被招 募，使熒光素酶報告基因處于激活狀態，加入其底物后發光信號增強。 Figure 1. CB1 β-Arrestin CHO 細胞模型原理圖 | |
